題目: Rational design for the use of cold atmospheric plasma in medicine
Abstract:
Life-threatening medical indications such as non-healing wounds and cancers cause suffering to patients and families, erode a patient’s quality of life, and are a social and economic burden. With ageing populations and greater disease incidences, it becomes paramount to develop more cost effective healthcare technologies which are appropriate to these diseases. Over the past two decades, atmospheric, ionized gas (plasma) devices have attracted significant attention worldwide for their applications in healthcare. And, there is significant optimism that plasma devices could deliver therapeutic agents in sufficient quantities for the sterilisation and regeneration of tissue in non-healing wounds and the selective destruction of cancerous cells within tissues. Current research points to a potential link between the plasma generation of reactive oxygen and nitrogen species (RONS) and outcomes in a range of biological and medical applications. As new data accrue, further strengthening this link, it becomes important to understand the controlled delivery of RONS into solutions, tissue fluids and tissues.
In this lecture, I will discuss simple experiments we have developed to assess the plasma delivery of RONS into tissue fluid, cells and tissue [1-3]. Our approach involves the use of chemical reporters and in situ UV-Vis spectroscopy to assess the plasma delivery of RONS into biologically relevant solutions, through tissue and into cells. We have shown that plasma readily delivers RONS deep within tissue at depths of millimetres and into cells without damaging the cell membrane. We have identified biological and chemical variables that can influence the plasma generation of RONS in biological systems such as the concentration of serum proteins and dissolved oxygen.
I will discuss how our approach can be utilised for the rational design and use of plasma in biology and medicine with particular focus on the treatment of wounds and cancers.
